Vaskuläre Immunobiologie
Sepsis, Thrombose und Atherosklerose sind schwerwiegende Erkrankungen, die sich im Gefäßsystem des Menschen abspielen. Es ist daher wichtig, die komplexen Mechanismen zu verstehen, die zur Aufrechterhaltung eines gesunden Gefäßsystems beitragen, aber auch die Pathophysiologie von Gefäßverletzungen, Entzündungen und Gefäßverschlüssen zu entschlüsseln. Eine entscheidende Rolle bei der Entstehung dieser Erkrankungen spielt das Immunsystem des Wirts. Zusätzlich haben wir einen weiteren wichtigen Akteur in der Gefäßimmunologie identifiziert: Thrombozyten. Diese Zellen modulieren die angeborene und adaptive Immunantwort bei akuten und chronischen Entzündungen und bieten möglicherweise neue therapeutische Zielstrukturen. Wir konnten zeigen, dass Thrombozyten im entzündeten Gefäßsystem migrieren, Gefäßverletzungen identifizieren und so zur vaskulären Integrität beitragen. Außerdem bilden Thrombozyten zusammen mit Monozyten und Neutrophilen intravaskuläre Thromben, um eindringende Bakterien einzufangen. Dieser Prozess wird als Immunthrombose bezeichnet und kann sich auch nachteilig auf den Wirt auswirken. Um diese komplexen Mechanismen der vaskulären Homöostase und Pathophysiologie weiter zu untersuchen, verwenden wir state-of-the-art Mausmodelle, Intravitalmikroskopie und in vitro-Funktionstests und kombinieren diese mit translationalen, patientennahen Untersuchungen.
